Each cell in a developing organism undergoes a defined sequence of developmental events to reach its final functional state. At decision points along their path to terminal differentiation, cells interpret a subset of the available instructions encoded by the genome to select the correct developmental trajectory. Molecularly, combinations of transcription factors initiate and stabilize these cellular decisions by activating context-specific gene expression program. Thus, understanding how transcription factors are regulated to generate diverse developmental outcomes is a fundamental challenge in biology.

(Cont.) First, we characterized the eye phenotypes arising from overexpression of nito. Overexpression of nito perturbs eye development and in contrast to spen, which has been shown to act as a positive effector of EGFR signaling, appears to be a negative effector of EGFR signaling during eye development. Further genetic analysis of the relationship between spen and nito in the eye suggests they function antagonistically, possibly by targeting a common set of genes for transcriptional regulation. Further analysis of SPOC family mutants and study of the functions of these proteins at a molecular level will be necessary to understand the relationship of small and large SPOC proteins and the roles of the proteins in development and disease.

1 Kevin Moses It is now 25 years since the study of the development of the compound eye in Drosophila really began with a classic paper (Ready et al. 1976). In 1864, August Weismann published a monograph on the development of Diptera and included some beautiful drawings of the developing imaginal discs (Weismann 1864). One of these is the first description of the third instar eye disc in which Weismann drew a vertical line separating a posterior domain that included a regular pattern of clustered cells from an anterior domain without such a pattern. Weismann suggested that these clusters were the precursors of the adult ommatidia and that the line marks the anterior edge of the eye. In his first suggestion he was absolutely correct - in his second he was wrong. The vertical line shown was not the anterior edge of the eye, but the anterior edge of a moving wave of patterning and cell type specification that 112 years later (1976) Ready, Hansen and Benzer would name the "morphogenetic furrow". While it is too late to hear from August Weismann, it is a particular pleasure to be able to include a chapter in this Volume from the first author of that 1976 paper: Don Ready! These past 25 years have seen an astonishing explosion in the study of the fly eye (see Fig.

The eye is a complex sensory organ, which enables visual perception of the world. Thus the eye has several tissues that do different tasks. One of the most basic aspects of eye function is the sensitivity of cells to light and its transduction though the optic nerve to the brain. Different organisms use different ways to achieve these tasks. In this sense, eye function becomes a very important evolutionary aspect as well. This book presents the different animal models that are commonly used for eye research and their uniqueness in evaluating different aspects of eye development, evolution, physiology and disease. Presents information on the major animal models used in eye research including invertebrates and vertebrates Provides researchers with information needed to choose between model organisms Includes an introductory chapter on the different types of eyes, stressing possible common molecular machinery

Stem cells have been gaining a lot of attention in recent years. Their unique potential to self-renew and differentiate has turned them into an attractive model for the study of basic biological questions such as cell division, replication, transcription, cell fate decisions, and more. With embryonic stem (ES) cells that can generate each cell type in the mammalian body and adult stem cells that are able to give rise to the cells within a given lineage, basic questions at different developmental stages can be addressed. Importantly, both adult and embryonic stem cells provide an excellent tool for cell therapy, making stem cell research ever more pertinent to regenerative medicine. As the title The Cell Biology of Stem Cells suggests, our book deals with multiple aspects of stem cell biology, ranging from their basic molecular characteristics to the in vivo stem cell trafficking of adult stem cells and the adult stem-cell niche, and ends with a visit to regeneration and cell fate reprogramming. In the first chapter, “Early embryonic cell fate decisions in the mouse”, Amy Ralson and Yojiro Yamanaka describe the mechanisms that support early developmental decisions in the mouse pre-implantation embryo and the current understanding of the source of the most immature stem cell types, which includes ES cells, trophoblast stem (TS) cells and extraembryonic endoderm stem (XEN) cells.

The fruit fly Drosophila melanogaster offers the most powerful means of studying embryonic development in eukaryotes. New information from many different organ systems has accumulated rapidly in the past decade. This monograph, written by the most distinguished workers in the field, is the most authoritative and comprehensive synthesis of Drosophila developmental biology available and emphasizes the insights gained by molecular and genetic analysis. In two volumes, it is a lavishly illustrated, elegantly designed reference work illustrating principles of genetic regulation of embryogenesis that may apply to other eukaryotes.

This volume focuses on pharmaceutical biotechnology as a key area of life sciences. The complete range of concepts, processes and technologies of biotechnology is applied in modern industrial pharmaceutical research, development and production. The results of genome sequencing and studies of biological-genetic function are combined with chemical, micro-electronic and microsystem technology to produce medical devices and diagnostic biochips. A multitude of biologically active molecules is expanded by additional novel structures created with newly arranged gene clusters and bio-catalytic chemical processes. New organisational structures in the co-operation of institutes, companies and networks enable faster knowledge and product development and immediate application of the results of research and process development. This book is the ideal source of information for scientists and engineers in research and development, for decision-makers in biotech, pharma and chemical corporations, as well as for research institutes, but also for founders of biotech companies and people working for venture capital corporations.

This exceptional laboratory manual describes thirty-seven procedures most likely to be used in the next decade for molecular, biochemical, and cellular studies on Drosophila. They were selected after extensive consultation with the research community and rigorously edited for clarity, uniformity, and conciseness. The methods included permit investigation of chromosomes, cell biology, molecular biology, genomes, biochemistry, and development. Each protocol includes the basic information needed by novices, with sufficient detail to be valuable to experienced investigators. Each method is carefully introduced and illustrated with figures, tables, illustrations, and examples of the data obtainable. The book's appendices include key aspects of Drosophila biology, essential solutions, buffers, and recipes. An evolution of Michael Ashburner's 1989 classic Drosophila: A Laboratory Manual, this book is an essential addition to the personal library of Drosophila investigators and an incomparable resource for other research groups with goals likely to require fly-based technical approaches.
